CHAMPAIGN, lll. New insights into a protein complex that regulates the very tips of chromosomes could improve methods of screening anti-cancer drugs.

Led by bioengineering professor Sua Myong, the research group's findings are published in the journal Structure.

Myong's group focused on understanding the proteins that protect and regulate telomeres, segments of repeating DNA units that cap the ends of chromosomes. Telomeres protect the important gene-coding sections of DNA from loss or damage, the genetic equivalent of aglets the covering at the tips of shoelaces that keep the ends of the laces from unraveling or fraying.

Telomeres play an important role in cell aging and death, since each time a cell divides, a little bit is lost from the end of the telomere. Thus, cell biologists postulate that telomere length can determine the lifespan of a cell. Cancer cells, however, have a way to get around this limitation: An enzyme called telomerase that adds length to telomeres is highly active in cancer cells. This allows cancer cells to divide in perpetuity, running amok through tissues and systems.

"Cancer researchers want to get a hold of this problem, control this indefinite lengthening of the telomeres," said Myong, who also is affiliated with the Institute for Genomic Biology at the U. of I. "A lot of the anti-cancer drugs are targeted directly to these telomeres so that they can inhibit telomerase activity. The proteins we study regulate the activity of telomerase."

Using a technique developed at Illinois that allows researchers to watch single molecules interact in real time, Myong's group determined how two proteins called POT-1 and TTP-1 bind to the telomere. POT-1 protects the fragile telomere ends from being attacked by other regulatory proteins that might mistake the end for a broken or damaged area of DNA. When POT-1 and TTP-1 work together in a complex, they promote telomerase activity, an interesting target for cancer researchers.

The group found that on its own, POT-1 binds to the folded-up telomere in distinct steps at particular points in the telomere's DNA sequence, unfolding the telomere in a stepwise manner. However, the POT-1/TTP-1 complex surprised the researchers by binding, then freely sliding back and forth along the telomere end.

"Instead of stepwise binding, what we saw was a mobile protein complex, a dynamic sliding motion," Myong said. "Somehow it was as if the static binding activity of POT-1 is completely lost the protein complex just slid back and forth. We were able to reproduce the data and confirm it with many different tail lengths of the telomeric DNA and we know now that the contact between POT-1 and the telomere is somehow altered when the partner protein comes and binds."

Next, the researchers will add telomerase and see how the sliding activity of the POT-1/TTP-1 complex affects telomerase activity. Myong postulates that the sliding may promote telomerase activity and thus telomere lengthening by making the end of the telomere accessible for the telomerase enzyme to bind.

"We are excited about the possibility that this kind of mobility can increase the telomerase extension activity," Myong said. "It's somehow engaging the enzyme so that it can stay bound to the DNA longer. So it must involve a direct interaction."

Ultimately, understanding the POT-1/TTP-1 complex gives drug developers a new target for anti-cancer drugs, and the assay Myong's group used to monitor the complex could offer a venue for evaluating telomere-targeting drugs.

"We want to extend our a basic science knowledge in telomere biology into causes of cancer and we hope that our assay can be useful for telomere-targeted drug screening," Myong said.

There's a whole host of techy tools that will let you log your 10k personal best, and break down your splits into minute detail, but what about poor old Rover? Fujistu spotted this gap, and created Wandant -- a sensor-laden neck tag for dogs complete with cloud-based service for tracking your pooch's wellbeing. The hardware will log your pet's footsteps, amount of shivering plus temperature change, and upload it automatically via your smartphone or PC. The software will present you these data over time, with the option of manually inputting further details such as food amount, a diary with photos and -- for those with stronger constitutions -- stool condition. Like human-focused equivalents, there will also be provisions for sharing your dog's efforts, but thankfully only with vets and research institutions. There's no details on hardware pricing just yet, but the online component is free for the first year (JPY 420 / about $5 per month there after). Money no object for your mutt? Sadly only available in Japan for the time being, so you'll need to include a return ticket in that splurge too.

AIDS fighting spirit flagging, will miss 2015 targets: charity

WASHINGTON (Reuters) - The world has lost momentum in the fight against the AIDS epidemic, with millions of new people infected last year, the ONE foundation said in a report given on Tuesday.

The conclusion contrasts with a UN report last week, which found that an end to AIDS was in sight due to better access to drugs.

But ONE says "the beginning of the end of the AIDS" remains out of reach, as the number of people newly infected each year still exceeds those who get treated for the first time.

Global leaders last year committed to the "beginning of the end of AIDS" by 2015.

Progress over the past decade has cut the death toll for the disease, mainly due to better access to drugs that can both treat and prevent the human immunodeficiency virus (HIV) which causes AIDS, the United Nations said in its annual report last week.

But while access to treatment has improved, in 2011 there were 2.5 million new cases of HIV. That is more than double the target of having only 1.1 million people newly infected each year, said ONE, a charity co-founded by Irish rock star Bono, that is dedicated to fighting poverty and preventable disease.

There were 34 million people living with AIDS at the end of last year.

At current rates, the world will not reach targets to reverse the spread of AIDS until 2022, seven years behind schedule, according to ONE.

"We recognize the world has done wonders in (fighting AIDS) in the last 10 years. But 2015 is around the corner," said Michael Elliott, ONE's chief executive.

"Here's a moment to put your pedal to the metal and go for it."

Much of the gap is due to funding cuts in major donor countries. The UN estimates there is about a $6 billion AIDS funding gap each year. Countries also have not coordinated a global strategy to tackle the AIDS epidemic, such as targeting treatment to groups at highest risk.

The ONE report examined funding for AIDS from the Group of Seven major developed countries, as well as the political leadership and strategy.

The United States has taken the lead in funding and tackling AIDS. President Barack Obama last year set a new target for AIDS treatments, and called on other nations to also boost their commitments.

The United Kingdom and France are not far behind, but Germany and Canada are lagging relative to their peers. And Japan and Italy are in the last two places, mainly due to Japan's earthquake in 2011 and Italy's economic woes.

Elliott said fiscal issues should not stop countries from donating to fight AIDS, since development aid is such a small portion of most countries' budgets -- an argument ONE has made consistently during the global recession.

"You have to be an unfeeling idiot, which we're not, to fail to recognize that the last few years have been tough economic times for people in many places all over the world," he said.

"(And) Italy may have fiscal problems. But it's not going to solve its fiscal problems on the back of development assistance."

But with major donors embroiled in austerity programs, low and middle-income countries have stepped up, and now provide more than half of the financing for AIDS, ONE said.

In the future, emerging economies like Brazil and China should take the lead on AIDS programs, ONE said.

"It's very important that people recognize this is a global fight," Elliott said. "It's not a fight that should be carried on the backs of the very generous American and British taxpayer.

The report, which comes ahead of World AIDS Day on December 1, will be posted online at www.one.org/policy.

Sen. Paul: Nation To Get "Big, Ugly" Debt Bag For Christmas

In what might loosely be described as the "spirit of the season," the Congress of the United States will likely put together a huge grab bag "with a lot of stuff in it," including tax- and debt-ceiling increases, and drop it down the nation's chimney before adjourning for its Christmas recess, Sen. Rand Paul (R-Ky.) predicted Monday.?

"I think there will be something really big, some enormous, ugly bill with a lot of stuff in it, including raising the debt ceiling by a couple trillion dollars. They'll squish it into one bill. And sometime before Christmas, they'll pass it," Paul said in an interview?with Greta Van Susteren on Fox News Monday night. But they'll pass it over his opposition, said Paul, a favorite of Tea Party activists and a potential contender for the GOP presidential nomination in 2016.

"I made a pledge to the people of Kentucky that I'm not raising taxes," said Paul, an ophthalmologist who became Kentucky's junior senator by winning an open seat in 2010 in his first political campaign. "I took a pledge. I signed a statement, an oath that I wouldn't raise taxes, and I'm going adhere to it," he said in apparent reference to the anti-tax pledge promoted by Grover Norquist, president of Americans for Tax Reform. Some key Republicans who took that pledge have indicated on the Sunday talk shows and other venues a willingness to consider tax increases, along with spending cuts, in an effort to avoid the much-discussed "fiscal cliff" of across-the-board spending cuts mandated by the budget agreement of 2011, and the tax increases that will occur without another extension of the Bush-era tax cuts, due to expire on January 1. Norquist responding on CNN's Starting Point,?dismissed talk of flexibility by anti-tax Republicans as "discussions of impure thoughts on national television." Paul made it clear he was entertaining no such thoughts.?

"I'm all for reforming the tax code, but it would have to be at the very best revenue-neutral. I don't want to raise revenue through the taxes," he said. "The only way I'd raise revenue through taxes is get the economy to grow. You don't have to raise rates or even close loopholes. The reason we have a lack of revenue in Washington is too much spending and no economic growth," he told Van Susteren. "If the economy were growing at 4 percent right now, we'd have plenty of revenue." Raising taxes is not the way to promote economic growth, he said. "That's what they want to do now, and I think it's absolutely the wrong thing to do."

Paul, the son of two-time Republican presidential hopeful Ron Paul,?appears ready to continue his father's decades-long campaign to reduce spending and taxes and oppose programs and actions of the federal government for which there is no constitutional grant of federal power or authority. The elder Paul, a 12-term congressman from Texas and the 1988 presidential candidate of the Libertarian Party, is retiring at the end of this year, having chosen not to run for another term in the U.S. House, where he has served, off and on, for more than 24 years over the course of nearly four decades. In the farewell address?he delivered on the House floor on November 14, the 77-year-old retired obstetrician warned of both a political and moral crisis brought on by "the exponential growth of illegal government power."

The younger Paul has often issued similar warnings about the growth of government and, unlike many of his colleagues in Congress, is not alarmed over the prospect of across-the-board spending cuts, mandated by the sequester provision of the 2011 budget agreement.??

"The sequester? Well, I think cutting spending's a good idea," he told Van Susteren. "So, I don't think that's a bad idea at all, and I don't think it will lead to economic harm to cut spending. That's what we should be doing," Paul blamed the stalemate in efforts to reach an agreement on taxes and spending on what he said is the Democrats' desire to increase both.

"So you can see why we're at somewhat of an impasse," he said. "We have different philosophies about government. I think you should balance budgets, not spend more than comes in, and I think you should lower taxes, not raise taxes."

Yet Paul often finds himself at odds with fellow Republicans by opposing foreign military interventions and overgrown military budgets, two of his father's favorite targets. In an interview with ABC News?last week Paul suggested military spending might be an area in which Democrats, who want more money for social programs, and fiscally conservative Republicans can find common ground.

"Republicans who think military spending, myself [included], who think national defense is important, should compromise and say, you know what, not every dollar spent on the military's sacred, we can reduce the military spending, that's a compromise. Democrats should compromise also ? entitlements and welfare, the spending can come down." And unlike the many Republicans who are staunch supporters of the war on drugs, Paul believes states such as Washington and Colorado that voted this month to legalize marijuana for adults should be free to do,?despite federal law banning the substance.??

"States should be allowed to make a lot of these decisions," Paul said last week. "I want things to be decided more at a local basis, with more compassion. I think it would make us as Republicans different." He also believes less draconian penalties for marijuana possession and use would help Republicans with younger voters. "I think for example we should tell young people, 'I'm not in favor of you smoking pot, but if you get caught smoking pot, I don't want to put you in jail for 20 years,'"??said Paul, who also parts company with hardliners on the issue of immigration reform.

While he maintains border security is a top priority, he has also expressed an openness to offering a path to citizenship to people who have been living here illegally for a long time. The image of the Republican Party as hostile to immigrants has hurt the GOP at the polls, he said. "We're getting an ever dwindling percent of the Hispanic vote," Paul noted. "We have to let people know, Hispanics in particular, we're not putting you on a bus and shipping you home." Paul, 49, is among the Republicans talked about as a potential presidential contender and he is not closing the door on a possible run for the White House in 2016." I won't deny that I'm interested in maybe running for president," he said Monday night. "But it's way too early to really make a firm commitment on, you know, whether I would or wouldn't."

Yes, I speak from experience? :) When Little Brother was still an infant I did everything in my power to not have to take my precious little guys with me to the craft store; unfortunately during that time Big Brother and I weren?t able to do nearly as many fun projects as we can today.?

PKM2 slips into nucleus to promote cancer; potential biomarker and drug approach discovered

HOUSTON - Researchers at The University of Texas MD Anderson Cancer Center have tracked down a cancer-promoting protein's pathway into the cell nucleus and discovered how, once there, it fires up a glucose metabolism pathway on which brain tumors thrive.

They also found a vital spot along the protein's journey that can be attacked with a type of drug not yet deployed against glioblastoma multiforme, the most common and lethal form of brain cancer. Published online by Nature Cell Biology, the paper further illuminates the importance of pyruvate kinase M2 (PKM2) in cancer development and progression.

"PKM2 is very active during infancy, when you want rapid cell growth, and eventually it turns off. Tumor cells turn PKM2 back on - it's overexpressed in many types of cancer," said Zhimin Lu, M.D., Ph.D., the paper's senior author and an associate professor in MD Anderson's Department of Neuro-Oncology.

Lu and colleagues showed earlier this year that PKM2 in the nucleus also activates a variety of genes involved in cell division. The latest paper shows how it triggers aerobic glycolysis, processing glucose into energy, also known as the Warburg effect, upon which many types of solid tumors rely to survive and grow.

"PKM2 must get to the nucleus to activate genes involved in cell proliferation and the Warburg effect," Lu said. "If we can keep it out of the nucleus, we can block both of those cancer-promoting pathways. PKM2 could be an Achilles' heel for cancer."

  • Activation of the MEK protein, which in turn activates ERK.
  • ERK sticking a phosphate group to a specific spot on PKM2.
  • Phosphorylation priming PKM2 for a series of steps that culminate in its binding to the protein importin, which lives up to its name by taking PKM2 through the nuclear membrane.

Once in the nucleus, the team showed that PKM2 activates two genes crucial to aerobic glycolysis and another that splices PKM RNA to make even more PKM2.

An experiment applying several kinase-inhibiting drugs to human glioblastoma cell lines showed that only a MEK/ERK inhibitor prevented EGF-induced smuggling of PKM2 into the nucleus. ERK activation then is mandatory for PKM2 to get into the nucleus.

"MEK/ERK inhibitors have not been tried yet in glioblastoma multiforme," Lu said. Phosporylated PKM2 is a potential biomarker to identify patients who are candidates for MEK/ERK inhibitors once those drugs are developed.

MEK inhibitor blocks tumor growth

The researchers also found that the two glycolysis genes activated by PKM2, called GLUT1 and LDHA, are required for glucose consumption and conversion of pyruvate to lactate, crucial factors in the Warburg Effect. Depleting PKM2 in tumor cell lines reduced glucose consumption and lactate production.

In mice, depleting PKM2 blocked the growth of brain tumors. Re-expressing the wild type protein caused tumors to grow. However, re-expression of a PKM2 mutant protein that lost its ability to get into the nucleus failed to promote tumor formation. Experiments in human glioblastoma cell lines showed the same effect.

Injecting the MEK inhibitor selumetinib into tumors inhibited tumor growth, reduced ERK phosphorylation, PKM2 expression and lactate production in mice. In 48 human tumor samples, the team found that activity of EGFR, ERK1/2 and PKM2 were strongly correlated.

Cause of PKM2 overexpression

Lu and colleagues also published a paper in Molecular Cell that revealed a mechanism for overexpression of PKM2 in glioblastoma. They found that EGF receptor activation turns on NF-?B, which leads to a series of events culminating in PKM2 gene activation.

PKM2 levels were measured in tumor samples from 55 glioblastoma patients treated with standard of care surgery, radiation and chemotherapy. The 20 with low PKM2 expression had a median survival of 34.5 months, compared to 13.6 months for the 35 patients with high levels of PKM2.

Level of PKM2 expression in 27 low-grade astrocytomas was about half of the expression found in higher grade glioblastomas.

"In these two papers, we show how PKM2 is overexpressed in tumors, how it gets into the nucleus, that nuclear entry is essential to tumor development, and identified potential drugs and a biomarker that could usefully treat people," Lu said.

PKM2 slips into nucleus to promote cancer; potential biomarker and drug approach discovered

HOUSTON - Researchers at The University of Texas MD Anderson Cancer Center have tracked down a cancer-promoting protein's pathway into the cell nucleus and discovered how, once there, it fires up a glucose metabolism pathway on which brain tumors thrive.

They also found a vital spot along the protein's journey that can be attacked with a type of drug not yet deployed against glioblastoma multiforme, the most common and lethal form of brain cancer. Published online by Nature Cell Biology, the paper further illuminates the importance of pyruvate kinase M2 (PKM2) in cancer development and progression.

"PKM2 is very active during infancy, when you want rapid cell growth, and eventually it turns off. Tumor cells turn PKM2 back on - it's overexpressed in many types of cancer," said Zhimin Lu, M.D., Ph.D., the paper's senior author and an associate professor in MD Anderson's Department of Neuro-Oncology.

Lu and colleagues showed earlier this year that PKM2 in the nucleus also activates a variety of genes involved in cell division. The latest paper shows how it triggers aerobic glycolysis, processing glucose into energy, also known as the Warburg effect, upon which many types of solid tumors rely to survive and grow.

"PKM2 must get to the nucleus to activate genes involved in cell proliferation and the Warburg effect," Lu said. "If we can keep it out of the nucleus, we can block both of those cancer-promoting pathways. PKM2 could be an Achilles' heel for cancer."

HANNOVER, Germany (AP) -A Hannover player is proving that there's still fair play in football.

Midfielder Lars Stindl has been suspended for a Bundesliga match after pointing out he should be banned for getting five yellow cards this season.

The 24-year-old Stindl kept count better than German football authorities who failed to register an earlier booking.

When Stindl was cautioned Saturday in a 5-0 loss against leader Bayern Munich, he told his club it should trigger a one-match ban.

Stindl says he wasn't tempted to exploit the official error, asking: "Why should we hide something?"

Hannover published the story on its website with the message: "Hats off, Lars!"

Coach Mirko Slomka praises Stindl as "an extremely fair sportsman" - who will watch the home match against struggling Greuther Fuerth from the sidelines.

Massachusetts gas explosion under investigation

SPRINGFIELD, Mass. (AP) ? Dozens of building inspectors began assessing homes and businesses in one of New England's biggest cities on Saturday, a day after a natural gas explosion leveled a strip club located next to a day care and heavily damaged a dozen other structures. The blast injured 18 people, many of them first responders.

Investigators were trying to figure out what caused the Friday night blast in Springfield that could be heard for miles, left a large hole in the ground where the multistory brick building housing Scores Gentleman's Club once stood and scattered debris over several blocks.

Officials had already evacuated part of the entertainment district after responding to a gas leak and odor reported about an hour before the explosion while firefighters, police officers and gas company workers were in the area filled with commercial properties and residences.

"It really is a miracle and it's an example of our public safety officials, each and every day, putting themselves in harm's way, taking what could have been considered a very routine call of an odor of gas, but they took the proper precautions," State Fire Marshal Stephen Coan said. "And thanks to God that they did."

Officials also marveled how the 5:30 p.m. blast occurred when a day care center next door was closed. The center's building was heavily damaged.

Lt. Gov. Tim Murray and Springfield Mayor Domenic Sarno were attending a tree-lighting ceremony when the explosion occurred. Sarno said some people mistakenly thought the boom was part of the holiday event.

The explosion blew out windows in a three-block radius, leaving at least three buildings irreparably damaged and causing emergency workers to evacuate a six-story apartment building that was buckling, police said. Pieces of broken glass littered streets and sidewalks. It was unclear how many residents had been evacuated. A shelter was set up at a school.

Omar Fermin, manager of the Punta Cana Restaurant two blocks from the explosion site, found the floor-to-ceiling windows blasted out when he came to check on the property Saturday morning.

"It looks like an earthquake hit," said Fermin, a native of the Dominican Republic. "I've never seen anything like it."

He said he was waiting for somebody to come and assess the damage. He worried the restaurant would remain closed for weeks while the owner seeks to replace the massive custom-made windows.

Authorities cordoned off the center of the explosion Saturday morning as building inspectors worked to identify unsafe structures. Anxious residents gathered at the perimeter, waiting for permission to visit their buildings.

Dogs trapped in abandoned buildings barked out loudly Saturday as building inspectors fanned out across the area. One inspector said he wished he could get a ladder and permission to retrieve a dog that was barking in the upper floor of one building sealed off from residents.

Coan, the fire marshal, said his office was investigating a cause of the blast and its possible origin. The state's Department of Public Utilities was also investigating.

Sheila Doiron, a spokeswoman for Columbia Gas of Massachusetts, said the company will continue to monitor for any potential leaks within several blocks of the blast site. So far, she said, the company hadn't yet found any measurable readings.

Doiron said the company also didn't find in its records any gas odor calls to the area where the strip club was located.

The victims were taken to two hospitals in the city. None of their injuries was considered life-threatening, officials said. Those injured were nine firefighters, two police officers, four Columbia Gas workers, two civilians and another city employee.

Springfield, which is 90 miles west of Boston and has about 150,000 residents, is the largest city in western Massachusetts. It's known as the home of the Basketball Hall of Fame, which is not in the vicinity of the blast.

The city has been rebuilding from damage it sustained in a June 2011 tornado.

On Friday night, residents milled around the neighborhood where the explosion occurred, stunned by the destruction and confused by the cordoned-off area, which grew as crews continued to search for gas leaks. The mayor warned against looting, saying police would be out in force.

Wayne Davis, who lives about a block away from the destroyed building, said he felt his apartment shake.

"I was laying down in bed, and I started feeling the building shaking and creaking," he said.

The Navy veteran said the boom from the explosion was louder than anything he'd ever heard, including the sound of a jet landing on an aircraft carrier.

The blast was so loud it was heard in several neighboring communities. Video from WWLP-TV showed the moment of the explosion, with smoke billowing into the air above the neighborhood.
